• ​​DECNUPAZ is a prescription medicine used to treat adults with blastic plasmacytoid dendritic cell neoplasm (BPDCN). It is not known if DECNUPAZ is safe and effective in children

DECNUPAZ may affect cancer cells as well as certain healthy cells in the body, which can cause side effects.

• ​DECNUPAZ is a type of medicine called an antibody-drug conjugate (ADC). ADCs are designed to attack cancer in a targeted way, which is different from traditional chemotherapies. DECNUPAZ works by targeting BPDCN cells that have a protein on them called CD123. ​CD123 is found on BPDCN cells and on other nearby cells

Understanding how DECNUPAZ is designed to work may help you feel more informed and confident throughout your treatment experience.

• ​You will receive DECNUPAZ as an IV infusion over ~15–30 minutes once every 21 days
• The day before each DECNUPAZ infusion, you will receive a medicine, known as a premedication, to help prepare your body for the infusion and try to prevent any infusion-related reactions that may occur
  • Some premedications will be given orally and some will be given as an IV infusion
• ​On the day of each DECNUPAZ infusion
  • You will receive premedications 30–60 minutes before each DECNUPAZ infusion, so you will have to arrive early
  • After each DECNUPAZ infusion, your healthcare provider will need to monitor you
    • You will be monitored for at least 4 hours after the first infusion
    • After each future infusion, you will be monitored for at least 1 hour

• The day before each DECNUPAZ infusion, you will receive a medicine, known as a premedication, to help prepare your body for the infusion and try to prevent any infusion-related reactions that may occur
  • Some premedications will be given orally and some will be given as an IV infusion
• ​On the day of each DECNUPAZ infusion
  • You will receive premedications 30–60 minutes before each DECNUPAZ infusion, so you will have to arrive early
  • After each DECNUPAZ infusion, your healthcare provider will need to monitor you
    • You will be monitored for at least 4 hours after the first infusion
    • After each future infusion, you will be monitored for at least 1 hour

• ​​​Your healthcare provider will decide how many infusions of DECNUPAZ you may receive. Talk to your healthcare provider about what your full treatment plan is and how long you may be on treatment

• ​​​You will be able to receive DECNUPAZ as an outpatient infusion. This means that you will not need to be admitted to or spend the night at a healthcare facility to receive treatment
• The day before each DECNUPAZ infusion, you will receive a medicine, known as a premedication, to help prepare your body for the infusion and try to prevent any infusion-related reactions that may occur
  • Some premedications will be given orally and some will be given as an IV infusion
• ​On the day of each DECNUPAZ infusion
  • You will receive premedications 30–60 minutes before each DECNUPAZ infusion, so you will have to arrive early
  • After each DECNUPAZ infusion, your healthcare provider will need to monitor you
    • You will be monitored for at least 4 hours after the first infusion
    • After each future infusion, you will be monitored for at least 1 hour

• ​​​​Your healthcare team will guide you through how they check your response to treatment. This may include blood work, a bone marrow test, measuring any skin changes, and doing CT or PET scans to look at your lymph nodes and organs. Together, these help your provider see if you're responding to treatment
• ​In a clinical trial, DECNUPAZ helped people with BPDCN achieve remission. Remission or complete remission means that all signs of cancer have disappeared in response to treatment, but there may be some skin abnormalities that remain
• In a clinical trial that, in part, included 33 people who received DECNUPAZ as their first treatment for BPDCN with no central nervous system involvement, 70% achieved complete remission. Of these people, 49% achieved complete remission with no skin abnormalities remaining and 21% achieved complete remission with some skin abnormalities remaining
• Of the people who achieved complete remission, half were estimated to still remain in remission for at least 9.7 months
• Of the 51 people who either relapsed after a previous treatment or did not respond to previous treatment (can include people with BPDCN with central nervous system involvement that was previously treated), 16% achieved complete remission. Of these people, 14% achieved complete remission with no skin abnormalities remaining and 2% achieved complete remission with some skin abnormalities remaining
• Of the people who achieved complete remission, half were estimated to still remain in remission for at least 9.2 months

DECNUPAZ can cause serious side effects, including:

• Liver problems (hepatotoxicity), including veno-occlusive disease (blockage of the small veins in the liver) that can be severe, life-threatening, or may lead to death. Your healthcare provider will do blood tests before each dose of DECNUPAZ and during treatment with DECNUPAZ to check for liver problems. Tell your healthcare provider right away if you develop signs or symptoms of liver problems, including:
  • yellowing of the skin or eyes
  • pain in your stomach (abdomen)
  • fast weight gain
  • swelling of your stomach
  • dark urine

Your healthcare provider will check you for liver problems during your treatment with DECNUPAZ and may provide treatment for your side effects. Your healthcare provider may also delay or stop treatment with DECNUPAZ if you have severe liver problems.

• Talk to your healthcare team right away about any discomfort or change in the way you are feeling. Getting medical treatment right away is important

• ​​Tell your healthcare provider about all of your medical conditions, including if you have liver problems, are allergic to sulfites, have asthma, have kidney problems, and/or are pregnant or plan to become pregnant. DECNUPAZ can harm your unborn baby
  • Females who are able to become pregnant:
    • ​Your healthcare provider will check for pregnancy before you start treatment with DECNUPAZ
    • Use effective birth control (contraception) during treatment with DECNUPAZ and for 7 months after your last dose
    • Tell your healthcare provider if you become pregnant or think that you may be pregnant during treatment with DECNUPAZ
  • Males who have female partners who are able to become pregnant:
    • Use an effective birth control during treatment with DECNUPAZ and for 4 months after your last dose of DECNUPAZ
• Tell your healthcare provider if you ​are breastfeeding or plan to breastfeed. It is not known if DECNUPAZ passes into your breast milk. Do not breastfeed during treatment with DECNUPAZ and for 1 month after your last dose
• Tell your healthcare provider about all the medicines you take, including prescription and over-the-counter medicines, vitamins, and herbal supplements
• Certain medicines may affect DECNUPAZ and increase your risk of side effects

• Serious side effects are possible and include: liver problems (hepatotoxicity), including veno-occlusive disease (blockage of the small veins in the liver) that can be severe, life-threatening, or may lead to death; infusion-related reactions; fluid retention (edema); and sulfite allergic reactions
• ​The most common side effects of DECNUPAZ include: fluid retention (edema); bleeding; feeling tired; infusion-related reactions; muscle, bone, and joint pain; nausea; and diarrhea
• The most common severe abnormal laboratory test results with DECNUPAZ include: decreased white blood cell counts, decreased platelet counts, decreased red blood cell counts, and increased blood sugar level
